Kligler Iron Agar enables simultaneous testing of glucose/lactose fermentation, gas production, and hydrogen sulfide formation in gram-negative bacteria. The standardized formulation requires 55.2g per liter of purified water, with optional sterilization at 118°C for 15 minutes when preparing slants for extended storage. Key performance characteristics include: distinct yellow butt (acid production) and red slant (alkaline reaction) differentiation, clear H2S visualization through black iron sulfide precipitation, and stable gel formation at 45°C for consistent slant angles. The medium supports reliable enteric pathogen differentiation within 24-48 hours incubation at 35±2°C.

Ideal for clinical microbiology, food safety labs, and water testing facilities, this medium detects critical biochemical reactions of Salmonella, Shigella, and other Enterobacteriaceae. The optimized formula prevents excessive H2S masking and maintains reaction stability for 7 days post-pouring. Available in dehydrated powder (500g containers) or pre-poured slants (5mL/tube, 20 tubes/rack), with quality verification including: pH 7.4±0.2 at 25°C, controlled sugar ratios (10:1 lactose/glucose), and ≤3% batch variation in color response. Typical results show 100% reactivity with control strains for acid/alkaline reactions and H2S production.
