This coliform-selective medium requires suspension of 39.0g powder in 980ml purified water with 20ml absolute ethanol. Critical preparation steps include vigorous boiling (>1 minute) for complete dissolution followed by sterilization at 115°C for 20 minutes. Post-sterilization shaking prevents agar sedimentation during cooling, ensuring homogeneous fuchsin distribution for consistent colony differentiation. The formulation generates characteristic pink-to-red colonies with metallic sheen for lactose-fermenting bacteria while suppressing gram-positive growth. Maintains optimal selectivity at pH 7.5±0.2 and demonstrates 24-hour stability for membrane filtration applications.

Designed for water quality analysis, dairy facility monitoring, and food processing hygiene control, this medium reliably identifies E. coli and other coliforms through distinct chromogenic reactions. The ethanol-enhanced formula intensifies metallic sheen development on lactose-positive colonies while clarifying background coloration. Validated for both pour-plate and membrane filtration methods with ≤5% variation in colony morphology across batches. Typical results show 100% sheen formation with ATCC 25922 controls and complete inhibition of non-target organisms. Available in dehydrated powder format (500g containers) with 18-month stability at controlled room temperature.
